Does Managed File Transfer supports restricting AD groups from accessing Managed File Transfer? Does Managed File Transfer supports restricting users of being shared with?
Yes, AD groups, or users belonging to AD groups can be restricted of being shared with a file - and that is done by denying their access to Managed File Transfer.
Denying access is done via the Managed File Transfer Settings / Active Directory / Configure and then in STAGE 5:
Include the Security groups (of users) which require access to Managed File Transfer.

Exclude the Security Group (of users) which should not be shared with.

Complete the setup.
